摘要:
目的研究LIM结构域蛋白1抗体在急性白血病患者的异常表达及相关性。方法选择首次确诊且未接受过治疗的急性白血病患者纳入初治组、接受化疗并达到完全缓解的白血病患者纳入缓解组、骨髓象正常的非恶性血液疾病患者纳入对照组,分离培养骨髓细胞,检测细胞中LIMD1,VEGF-C,CTGF和Survivin的mRNA含量及细胞增殖能力。结果LIMD1的mRNA含量在三组中从小到大顺序为初治组(14.52±1.96)mg,缓解组(76.67±9.56)mg和对照组(111.27±16.45)mg,差异均有统计学意义(P<0.05);MTT值在三组中从小到大顺序为对照组(100±15.34),缓解组(151.77±19.34)和初治组(314.45±52.28),差异均有统计学意义(P<0.05);VEGF-C的mRNA含量在三组中从小到大顺序为对照组(104.78±15.14)mg,缓解组(146.63±18.84)mg和初治组(259.34±31.45)mg,差异均有统计学意义(P<0.05);CTGF的mRNA含量在三组中从小到大顺序为对照组(99.27±16.67)mg,缓解组(162.15±19.34)mg和初治组(325.56±36.72)mg,差异均有统计学意义(P<0.05);Survivin的mRNA含量在三组中从小到大顺序为对照组(109.44±15.78)mg,缓解组(134.39±16.67)mg和初治组(277.74±29.34)mg,差异均有统计学意义(P<0.05);LIMD1与增殖能力以及VEGF-C,CTGF,Survivin的mRNA含量呈负相关(β=0.679~0.795,P<0.05)。结论LIMD1在急性白血病患者骨髓细胞中的表达减少,且与细胞增殖以及恶性生物学分子VEGF-C,CTGF和Survivin的表达具有相关性。
Abstract:
ObjectiveTo study the abnormal expression of LIM domain-containing protein 1 antibody in acute leukemia patients and its relationship.MethodsFirst diagnosed with acute leukemia patients who did not receive treatment were enrolled in initial treatment group,leukemia patients received chemotherapy and achieve complete remission were enrolled in remission group,non-malignant blood diseases patients with bone marrow normal were enrolled in control group.Then bone marrow cells were isolated and cultured,LIMD1,VEGF-C,CTGF and Survivin mRNA contents and cell proliferation were detected.ResultsFrom the lowest to the highest,the LIMD1 mRNA levels in three groups were initial treatment group (14.52±1.96)mg,remission group (76.67±9.56)mg and control group (111.27±16.45)mg.From the lowest to the highest,the MTT levels in three groups were control group (100±15.34),remission group (151.77±19.34) and initial treatment group (314.45±52.28).From the lowest to the highest,the VEGF-C mRNA levels in three groups were control group (104.78±15.14)mg,remission group (146.63±18.84)mg and initial treatment group (259.34±31.45)mg.From the lowest to the highest,the CTGF mRNA levels in three groups were control group (99.27±16.67)mg,remission group (162.15±19.34)mg and initial treatment group (325.56±36.72)mg.From the lowest to the highest,the Survivin mRNA levels in three groups were control group (109.44±15.78)mg,remission group (134.39±16.67)mg,and initial treatment group (277.74±29.34)mg,LIMD1 was negatively correlated with proliferation and mRNA contents of VEGF-C,CTGF,Survivin (β=0.679~0.795,P all<0.05).ConclusionThe expression LIMD1 in patients with acute leukemia cells decreases and correlates with cell proliferation and the expression of malignant molecular VEGF-C,CTGF,Survivin.
