摘要:
目的 研究IL-23p19在原发性胆汁性肝硬化(PBC)外周血单个核细胞中的表达及意义。方法 采用实时荧光定量逆转录-聚合酶链反应(RT-PCR)技术检测了60例PBC患者及60例疾病对照和60例健康对照外周血单个核细胞(PBMC)中IL-23p19基因表达量,应用统计学软件分析PBC IL-23p19基因表达与疾病分期及肝功能指标间的相关性。结果 PBC患者外周血PBMC中IL-23p19基因表达较健康对照和疾病对照均明显增高,且在疾病的I/II期表达高于III/IV期; IL-23p19表达与谷氨酰转肽酶(GGT)正相关。结论 IL-23p19基因在PBC中表达升高,通过促炎反应参与PBC发病。
Abstract:
Objective To investigate the expression of IL-23p19 in perpheral blood of patients with primary biliary cirrhosis(PBC)and itsclinical significance.Methods The real-time quantitative reverse transcription-polymerase chain reaction(RT-PCR)was used to determine the expression of IL-23p19 mRNA in the peripheral blood of 60 PBC patients,60 healthy controls,and 60 patients with hepatitis related cirrhosis.The correlation btween the expression of IL-23p19mRNA with the staging and hepatic functions of PBC patients was analyzed statistically.Results The IL-23p19 mRNA level in PBMC of PBC was significantly higher than those inhealty controls and hepatitis related cirrhosis.We also found that the IL-23p19 mRNA level in PBC patients atⅠ,Ⅱ stages were significantly higher than those at Ⅲ,Ⅳ stages.The IL-23p19 mRNA level in PBC patients were positively correlated with the serum gamma glutamyltransferase level.ConclusionThe IL-23p19 gene expression of PBMC in PBC patients was significantly elevated,indicating that IL-23p19 play an important role in the pathogenesis of PBC by promoting inflammation.
