摘要:
目的 建立一种操作简便、结果稳定的即时性中性粒细胞碱性磷酸酶(neutrophil alkaline phosphatase,NAP)染色方法。方法 参照国内常规方法,在染料及配制、染色步骤及结果判断等方面进行改良,称为改良方法; 统计181例临床患者和健康体检者,根据NAP活性在疾病中的分布特点分为低水平组、中水平组、高水平组,然后分别用两种方法进行染色,记录各组阳性率和积分值,观察改良方法与常规方法在染色效果和染色结果上有无差异。结果 改良方法阳性颗粒为深蓝色,不分散,易辨别,确定了可信性; 3组临床资料中阳性率和积分值比较差异均无统计学意义,并两种染色结果呈正相关,即低水平组阳性率(t=1.31,P=0.20>0.05,r=0.94)、中水平组阳性率(t=-0.11,P=0.91>0.05,r=0.66)和高水平组阳性率(t=1.22,P=0.23>0.05,r=0.82); 低水平组积分值(t=0.44,P=0.66>0.05,r=0.95)、中水平组积分值(t=1.30,P=0.19>0.05,r=0.97)、高水平组积分值(t=-0.78,P=0.44>0.05,r=0.90)。结论 改良方法省时、简便,实验室容易接受,能为临床提供及时可靠的实验依据。
Abstract:
Objective To establish a simple,stable instantresults neutrophil alkaline phosphatase(NAP)staining methods.Methods According to domestic conventional methods,made improvements inthe preparation of dyes and staining procedures and the results of judgment,etc,and called the modified method.Statistics 181 cases of clinical patients andhealthy subjects,according to the NAP activity in disease distribution groups into low,the group level,a high level group,and two methods were used to stainpositive rates in each group recorded and the integral value,and improved method of the conventional method to observe whether the differences in the dyeing effect and staining.Results The positive result of improved methods of particle dark blue,not scattered,easy to identify,determine credibility.Showed no difference in the positive rate and the integral value of clinical data in the three groups was statistically and two staining results werepositively correlated,low-positive group rate(t=1.31,P=0.20>0.05,r=0.94),the positive rate level group(t=-0.11,P=0.91>0.05,r=0.66),thepositive rate of high-level group(t=1.22,P=0.23>0.05,r=0.82).Low set points value(t=0.44,P=0.66>0.05,r=0.95),the integral value of thelevel of the group(t=1.30,P=0.19>0.05,r=0.97),a high level group ofintegral value(t=-0.78,P=0.44>0.05,r=0.90).ConclusionThe improved method of saving,simple,easy to accept the laboratory,it could provide timely and reliable experimental basis for clinical application.
