摘要:
目的 检测原发性胆汁性肝硬化(PBC)患者外周血Tim-1,IFN-γ和IL-4的表达水平,分析其相关性,初步探讨Tim-1在PBC诊治中的临床意义。方法 收集2013年6月~2015年11月来长海医院就诊的32例PBC患者和32例同期就诊的健康成人的外周血,分离外周血单个核细胞(PBMC),反转录酶-聚合酶链锁反应(RT-PCR)法检测PBMC中Tim-1 mRNA,酶联免疫吸附试验(ELISA)法检测血浆中Tim-1蛋白表达量,对Tim-1和IFN-γ,IL-4之间的关系进行Pearson相关性分析。结果患者组和对照组Tim-1 mRNA分别为0.63±0.40和0.36±0.32,差异具有统计学意义(t=3.005,P=0.003,P<0.01)。患者组与对照组Tim-1蛋白表达水平分别为62.72±38.60和35.66±30.55 pg/ml(t=3.11,P=0.002 8),IFN-γ表达水平分别为200.81±18.06和79.23±20.02 pg/ml(t=25.51,P<0.000 1),IL-4表达水平分别为99.22±14.67和30.18±20.56 pg/ml,差异均具有统计学意义(t=15.46,P<0.000 1)。Pearson相关分析结果提示Tim-1与IFN-γ呈负相关(r=-0.552,P=0.001 1),与IL-4呈正相关(r=0.652,P<0.000 1)。结论Tim-1可能参与了PBC的发病机制,是该病潜在的标志物和治疗靶点。
Abstract:
Objective To test the expression level of Tim-1,IFN-γ,IL-4 in peripheral blood of patients with primary biliary cirrhosis,analyze their correlation and preliminarily explore the clinical significance of Tim-1.Methods The peripheral blood were collectedfrom 32 patients with PBC in June,2013 to December,2015 and 32 health individuals at the same time.PBMC was separated from the peripheral blood,mRNA expression of Tim-1 in PBMC was measured by reverse transcriptase-polymerase chain reaction(RT-PCR).Tim-1 protein existed in plasma was tested by enzyme-linkedimmunosorbent assay(ELISA).Pearson correlation analysis could be done to determine the relationship between Tim-1 and IFN-γ,IL-4.Results The mRNA level of Tim-1 in PBMC of patient and control groups were 0.63±0.40,0.36±0.32 separately,(t=3.005,P=0.003)with significant statistical difference(P<0.01).Expression levels of Tim-1 protein in patient and control groups were 62.72±38.60,35.66±30.55 pg/ml separately(t=3.11,P=0.002 8),expression levels of IFN-γ were 200.81±18.06,79.23±20.02 pg/ml separately(t=25.51,P<0.000 1),that of IL-4 were 99.22±14.67,30.18±20.56 pg/ml separately(t=15.46,P<0.000 1),the differences were all with statistical meaning.The results of pearson correlation analysis suggested that Tim-1 in plasma was negatively related with IFN-γ(r=-0.552,P=0.001 1)and positively with IL-4(r=0.652,P<0.000 1).Conclusion Tim-1 may be involved in pathogenesis of PBC,itcan be the potential biomarker and therapeutic target of this disease.
