摘要:
目的 检测肺癌患者miR-142-5p的表达变化情况,探讨其在肺癌早期诊断的临床价值。方法 收集2014年4月~10月55例I,II期肺癌患者及53例健康成人对照血浆标本,采用实时荧光定量PCR(RT-qPCR)检测血浆中miR-142-5p的表达水平,组间比较采用独立样本t检验,运用ROC曲线评价miR-142-5p对肺癌早期诊断方面的应用价值。结果 肺癌组血浆中miR-142-5p相对表达量为8.878 2±2.251 2,健康成人组相对表达量为12.000 0±3.694 4,肺癌组高于健康成人组,差异有统计学意义(t=-5.325,F=13.561,P<0.01),同时测得差异表达倍数(fold change,FC)值为8.70; ROC曲线结果显示,miR-142-5p作为肺癌早期诊断指标具有较好诊断效能(ROC曲线下面积为0.748),最佳诊断临界值为10.050,敏感度为66.0%,特异度为83.6%。结论 miR-142-5p在肺癌患者血浆中存在高表达,其在肺癌早期诊断中具有良好的临床应用前景。
Abstract:
Objective To analyze the change of miR-142-5pexpression in lung cancer patients,and to explore its clinical value in earlydiagnosis of lung cancer.Methods Collected in April 2014 to October 55 cases of phase I,II lung cancer patients and 53 healthy adult control plasma samples,using real-time quantitative PCR(RT-qPCR)to detect the expression levels of miR-142-5p in plasma,between groups comparing with independent sample t test,using ROC curve in the evaluation miR-142-5p valueof early diagnosis of lung cancer.Results Plasma miR-142-5p in lung cancer group relative expression was 8.878 2±2.251 2.Healthyadult group relative expression was 12.000 0±3.694 4.Lung cancer group was higher than that of healthy adult group,and the difference was statistically significant(t=-5.325,F=13.561,P<0.01).While the measured FC(fold change)value was 8.704 7.miR-142-5p as indexes for early diagnosis of lung cancer had a good diagnostic performance(the area under the ROC curve was 0.748),10.050 of optimal diagnostic critical value,66.0% of sensitivity,and 83.6% of specificity.Conclusion The high expressionof miR-142-5p in the plasma of lung cancer patients has good clinical application prospects in the early diagnosis of lung cancer.
