摘要:
目的 检测巨噬细胞迁移抑制因子(MIF)在隐球菌脑膜炎患者外周血单个核细胞(PBMCs)中的表达水平,进一步探讨其临床意义。方法收集2012年8月~2015年11月到上海长海医院确诊的42例新型隐球菌脑膜炎患者及同期体检的42例健康个体外周血,以密度梯度离心法分离PBMCs,以PCR方法检测PBMCs中MIF的mRNA相对表达量,以ELISA法检测血浆中MIF,IL-17,IL-1β,TNF-α,IFN-γ和IL-4水平,两组间细胞因子表达的比较采用两独立样本t检验,MIF与细胞因子间的关系以Pearson相关系数表示。结果 MIF在实验组与对照组的蛋白表达水平为34.17±7.88 ng/ml vs 10.89±2.76 ng/ml(t=18.07,P<0.0001); mRNA相对表达量为2.87±0.94 vs 1.95±0.89(t=4.606,P<0.0001),差异具有统计学意义(P<0.05)。Pearson相关性分析显示实验组MIF与IL-1β,IL-17呈正相关(r=0.467,0.401,P<0.01),差异具有统计学意义。结论 MIF可能通过影响IL-1β和IL-17等细胞因子的分泌及功能参与了隐球菌脑膜炎的免疫调节过程,是该病潜在的靶位点和疾病监测指标。
Abstract:
Objective To test the expression of MIF in peripheral blood mononuclear cells(PBMCs)from patients with Crytococcal Meningitisand further discuss its clinical significance.Methods Peripheral blood from 42 patients with Crytococcal Meningitis diagnosed in Changhai Hospital,Shanghai and 42 healthy individuals examined at the same time was collected from August,2012 to November,2015.PBMCs were separated by densitygradient centrifugation method,mRNA relative expression of MIF in PBMCs was measured by PCR,the level of MIF,IL-17,IL-1β,TNF-α,IFN-γ and IL-4 in plasma was tested by Enzyme-linked immunosorbent assay(ELISA).The comparison for expression level of cytokines between the two groups was by two-independentsamples t test.Pearson correlation coefficient was used to measure the relation between MIF and other cytokines.Results The proteinlevels of MIF in experimental and controlled groups were 34.17±7.88 ng/ml vs 10.89±2.76 ng/ml(t=18.07,P<0.0001),while relative expression of RNAwas 2.87±0.94 vs 1.95±0.89(t=4.606,P<0.0001),and there was statistical significance(P<0.005).Pearson correlation analysis showed that MIF was positively related with IL-1β,IL-17(r=0.467,0.401,P<0.01),withstatisticaldifference.Conclusion MIF may involve in theimmune regulation for Crytococcal Meningitis by affecting the secretion and function of cytokines as IL-1β,IL-17,and it was potential target and monitoredbiomarker for this disease.
