摘要:
目的 研究分泌型磷脂酶A2-IIA(sPLA2-IIA)的表达与寻常型银屑病皮损发病的关系。方法 依据PASI评分将寻常型银屑病患者50例分为3组,轻度组16例,中度组18例,重度组16例。对照组52例。酶联免疫吸附法检测银屑病患者血清sPLA2-IIA含量。逆转录聚合酶链式反应(RT-PCR)检测银屑病患者皮损组织和对照组皮肤标本中sPLA2各亚型mRNA表达水平。Western blot检测Akt及其磷酸化抗体的表达水平。结果 对照组血清sPLA2-ⅡA含量84.55±76.22 pg/ml,寻常型银屑病患者血清sPLA2-ⅡA含量265.31±54.23 pg/ml,两组差异有统计学意义(t=13.62,P<0.01)。银屑病皮损组织sPLA2-IIA的mRNA表达水平显著升高(t=113.41,P<0.01),而其它sPLA2亚型表达无变化。银屑病患者皮损重度组血清sPLA2-ⅡA含量明显比中度组和轻度组高,中度组比轻度组高,3组两两比较差异均有统计学意义(F=69.62,t=3.14,5.14,6.38,均P<0.01)。3组间sPLA2-ⅡA的mRNA表达两两比较差异也有统计学意义(F=28.12,t=4.77,10.42,10.58,均P<0.01)。PASI评分越高,血清sPLA2-ⅡA含量和sPLA2-ⅡA的mRNA表达越高。寻常型银屑病患者皮损组织Akt和p-Akt表达升高(1.87±0.31,1.85±0.28),与对照组相比差异有统计学意义(t=17.79,19.04,均P<0.01)。结论 sPLA2-IIA表达与寻常型银屑病皮损严重程度有关,其作用通路与Akt及其磷酸化表达水平相联系。
Abstract:
Objective To discuss the relationship betweenthe expression of secretory Phospholipase A2-ⅡA(sPLA2-IIA)and the pathogenesis psoriasis vulgaris patients lesions.Methods Using Psoriasis Area Severity Index(PASI),50 psoriasis vulgaris were divided into threegroups:mild(n=16),moderate(n=18)and severe group(n=16),and compared to the non-psoriasis control group(n=52).The serum level of sPLA2-IIA in psoriasis vulgaris patients and the normal control group were detectedusing ELISA methods.Using RT-PCR,mRNA expression levels of sPLA2-IIA and subtypes were detected.Using western blot,the expression levels of Akt and p-Akt were detected.Results The serum level of sPLA2-IIA in the psoriasis lesion's group was significantly higher than that in the healthy control(t=13.62,P<0.01).The mRNA expression of sPLA2-IIA in the lesion of psoriasis was significantly higher than that in health control(t=113.41,P<0.01).In the other subtypes,the mRNA expression of sPLA2 had no distinctive.Among the three groups(mild group,moderate group and severe group),thedifference in serum level and mRNA expression of sPLA2-IIA were statisticallysignificant(F=28.12,69.62,P<0.01).There was statistical significanceamong three groups(t=3.14,5.14,6.38,all P<0.01; t=4.77,10.42,10.58,all P<0.01).The expression of Akt/p-Akt in the psoriasis lesion's group were higher than that in health control(t=17.79,19.04,P<0.01).Conclusion The expression of sPLA2-IIA would be associated with order of severity in lesions of psoriasis vulgaris.The pathway was relate to expression of Akt and p-Akt.
