摘要:
目的 探究T细胞免疫球蛋白黏蛋白3(Tim-3)在1型糖尿病(T1DM)患者中的表达情况及其在T1DM发病过程中可能的作用。方法 流式细胞术检测32例T1DM患者与26例健康对照者Tim-3+细胞比例; Real Time-qPCR方法检测外周血单个核细胞(PBMC)中Tim-3 mRNA表达水平,同时检测糖化血红蛋白(HbA1C),胰岛素自身抗体(ICA)和谷氨酸脱羧酶65(GAD65)等临床相关指标。结果 T1DM患者Tim-3+细胞比例为7.91±4.35,明显高于对照组1.73±1.77; Tim-3 mRNA相对含量0.80±0.06,明显高于对照组相应检测值10.25±0.04,各项检测指标两组之间比较差异均有统计学意义(P<0.05); Tim-3 mRNA水平与T1DM患者GAD65呈显著正相关性,与HbA1C及ICA无显著相关性。此外,Tim-3+细胞比例与T1DM患者HbA1C及GAD65均呈显著正相关性,与ICA无显著相关性。结论 Tim-3在T1DM患者中表达明显上升,并与相关临床症状相关,提示Tim-3在T1DM发生发展中发挥一定的作用。
Abstract:
Abstract:Objective To explore the expression of T cell immunoglobulin mucin domain containing molecules 3(Tim-3)and its function in the type 1 diabetes mellitus(T1DM).Methods Flow cytometry was performed to measure the ratio of Tim-3+cells in 32 patients with T1DM and 26 healthy controls.The expression of Tim-3 in the peripheral blood monomuclear cells(PBMCs)was detected by real-time qPCR,and levels of HbA1C,ICA andGAD65 were also recorded.Results The ratio of Tim-3+ cells in patients with T1DM was significantly higher than that in healthy controls(P<0.01).The expression of Tim-3 mRNA in PBMCs from T1DM patients was also increased.Moreover,the level of Tim-3 mRNA was positively correlated withGAD65,but it had no association to HbA1C and ICA.Furthermore,the ratio of Tim-3+ cells was positively correlated with HbA1C and GAD65,and no relationship was observed between Tim-3+ cells and ICA.Conclusion Tim-3 is highly expressed in the T1DM patients and is related to clinical symptoms,indicating Tim-3 might play a role during the occurrence and development of T1DM.
