目的 研究热损伤/热习服大鼠血清中microRNA-193a-3p表达的变化及其与热休克蛋白(HSP70)、白介素-1β(IL-1β)、肿瘤坏死因子(TNF-α)和转化生长因子-β((TGF-β)的相关性。方法 随机选取45只体重、肛温、周龄基本一致的雄性大鼠并分为3组:热休克损伤组(HS)、热习服组(HA)和对照组(HC)。在高温实验舱中建立热休克损伤和热习服大鼠模型,建模成功后采集大鼠心尖血并离心得到血清。通过实时荧光定量PCR(qRT-PCR)检测大鼠血清中的microRNA-193a-3p的水平,采用酶联免疫吸附法(ELISA)检测血清中HSP70和相关细胞因子IL-1β的蛋白水平。实验组与对照组之间的比较采用Dunnett-t检验,用Pearson相关性分析来比较HA组和HS组两变量之间的相关性。结果 MicroRNA-193a-3p在三组中的表达量分别为0.99±0.30(HS),1.13±0.33(HA)和2.15±0.35(HC),其在HA和HS组中的表达显著降低(P<0.05)。在HS组中,microRNA-193a-3p与HSP70,IL-1β和TNF-α呈负相关,差异具有统计学意义(r=-0.821,-0.839,-0.578,P<0.05)。在HA组,microRNA-193a-3p与HSP70,IL-1β呈负相关,且具有统计学意义(r=-0.770,-0.766,P<0.05)。结论 MicroRNA193a-3p在热损伤/热习服大鼠血清中表达减少,且可能参与热损伤/热习服的病理过程。
Abstract:
Abstract:Objective To investigate the changes of microRNA-193a-3p in serum of rats with heat shock/heat acclimation and its correlation with HSP70,IL-1β,TNF-α and TGF-β.Methods 45 male rats which had the same body weight,anal temperature and age were divided randomly into 3 groups:heat shock(HS),heat acclimatization(HA)and health control(HC).Rat models with heat shock and heat acclimatization was established inthe high temperature experimental chambers.Once the rat models established successfully,the apical blood of rats was collected and centrifuged to obtain theserum.MicroRNA-193a-3p in rats serum were measured by real-time fluorescentquantitative PCR(qRT-PCR).HSP70 and related cytokines were detected by ELISA.The comparison between experimental group and control group was performed by Dunnett-t test.Used Pearson correlation analysis to compare the correlationbetween the two variables in HA group and HS group.Results The expression of MicroRNA-193a-3p in three groups was 0.99±0.3(HS),1.13±0.33(HA)and 2.15±0.35(HC),respectively.The expression of MicroRNA-193a-3p in HA and HS groups was significantly decreased(P<0.05).MicroRNA-193a-3p was negatively correlated with HSP70,IL-1β and TNF-α in HS group,and the correlation was statistical significance(r=-0.821,-0.839,-0.578,all P<0.05).MicroRNA-193a-3p was negatively correlated with HSP 70 and IL-1β in HA group,and the correlation was statistical significance(r=-0.770,-0.766,all P<0.05).Conclusion MicroRNA-193a-3p expression decreased in serum of rats with heat injure/heat acclimatization,which may be involved in the pathological process of heat injure/heat acclimatization.
