摘要:
目的 对2型糖尿病(type 2 diabetes mellitus,T2DM)患者外周血血清miRNA差异表达分析,为筛选出T2DM发生发展有关的miRNA分子标记物奠定基础,从而为T2DM预防、早期诊断和治疗方案提供指导。方法 采用GeneChipTM miRNA 4.0 Array筛选3组受试者外周血血清(T2DM组、T1DM组、对照组)的差异miRNA,并对差异miRNA进行靶基因预测和靶基因信号通路富集分析(KEGG)。结果 T2DM组分别与对照组,T1DM组相比,hsa-miR-505-3p和hsa-miR-548an均上调,hsa-miR-296-3p,hsa-miR-3160-5p,hsa-miR-33a-5p,hsa-miR-572和hsa-miR-96-5p均下调。通过靶基因预测和Pathway分析,筛选出5个miRNA(hsa-miR-505-3p,hsa-miR-548an,hsa-miR-296-3p,hsa-miR-3160-5p,hsa-miR-33a-5p,hsa-miR-572和hsa-miR-96-5p)、7个基因(MTOR,SLC2A2,PRKCE,IRS2,IRS1,MAPK8,MAPK10)可能与2型糖尿病胰岛素抵抗相关。结论 该研究筛选出的5个miRNA和7个基因,可为2型糖尿病胰岛素抵抗的研究提供理论基础。
Abstract:
Objective Through analyzing the differential expression of serum miRNA in peripheral blood of type 2 diabetes mellitus(T2DM)patients to lay a foundation for screening out miRNA molecular markers related tothe occurrence and development of T2DM,so as to provide guidance for prevention,early diagnosis and treatment of T2DM.Methods Used GeneChipTM miRNA4.0 Array to screen differential miRNA expression of threegroups(T2DM group,the T1DM group,control group)peripheral blood serum,thenpredicted target gene,analyzed the target genes by KEGG pathway analysis.Results Compared with the control group and the T1DM group,both hsa-miR-505-3p and hsa-miR-548an were up-regulated in the T2DM group,while hsa-miR-296-3p,hsa-miR-3160-5p,hsa-miR-33a-5p,hsa-miR-572 andhsa-miR-96-5p were down-regulated.Through target gene prediction and pathway analysis,5 miRNA(hsa-mir-505-3p,hsa-mir-548an,hsa-mir-3160-5p,hsa-miR-96-5p,hsa-mir-33a-5p)and 7 genes(MTOR,SLC2A2,PRKCE,IRS2,IRS1,MAPK8,MAPK10)were screened associated with insulin resistance in type 2 diabetes possibly.Conclusion Five miRNAs and seven genes were screened out,can provide a theoretical basis for the study of insulin resistance in type 2 diabetes.
