摘要:
目的 探讨不同酸类酸化的伊红溶液在活检小标本预染色中的染色效果。方法 从中国人民解放军总医院病理科 日常收取的病理标本中随机选取直径小于0.5cm 的胃肠镜组织和穿刺组织共计90 例,随机均分成3 组,每组均30 例, 所有标本取材均使用相同规格的三角滤纸包埋组织防止漏检,分别滴加2~3 滴冰醋酸化伊红溶液、盐酸化伊红溶液和 水溶性伊红溶液进行预染色,相同机器及程序进行脱水包埋,制片染色流程,肉眼和显微镜下分别观察组织蜡块及镜下 组织染色效果,按染色评分标准进行评分。结果 肉眼观察组织蜡块,水溶性伊红溶液组滴染组织着色较淡,不易识别, 包埋后组织与石蜡对比不明显,冰醋酸化伊红溶液组和盐酸化伊红溶液组滴染组织着色均较深,易识别,包埋后组织与 石蜡对比明显,冰醋酸化伊红溶液和盐酸化伊红溶液组比较差异无统计学意义(χ2=0.215,P >0.05),两组酸化伊红溶液 组与水溶性伊红溶液组比较差异均有统计学意义(χ2=5.221 ~ 6.923,均P <0.05);显微镜下观察,三组染色切片的结 构清晰,核质对比分明,没有明显差异。醋酸酸化伊红溶液较盐酸酸化伊红溶液配制简单,时间短。结论 醋酸酸化伊 红溶液对活检小标本的预染效果优于水溶性伊红溶液的预染效果,且配制方法优于盐酸酸化伊红溶液,在包埋过程中易 于识别,能提高包埋速度和质量,值得推广。
Abstract:
Objective To investigate the effect of eosin solution acidified with different acid solution on prestaining of small biopsy specimens.Methods A total of 90 cases of gastrointestinal endoscopy and puncture tissues less than 0.5 cm in diameter were randomly selected from pathological specimens collected by the Pathology Department of the General Hospital of the People’s Liberation Army of China. They were randomly divided into three groups, each group was divided into 30 cases. All specimens were taken with triangular filter paper of the same specifications to prevent tissue from being missed. Then 2~3 drops of glacial acetated eosin solution, hydrochloric acid eosin solution and water-soluble eosin solution were added for pre-dyeing. The same machine and procedure were used for tissue dehydration, tissue embedding, slicing and dyeing process. The effect of tissue wax and microscopic tissue dyeing was observed under naked eye and microscope respectively. The color grading standard was used for grading.Results By macroscopic observation group wax block, the drip dyeing of water-soluble eosin solution group was light and difficult to identify, and group compared and salt solution group. All drop staining of tissue deep, it was easy to identify, after embedding tissue compared with paraffin, glacial acetic acid acidification eosin eosin solution and salt solution group of comparison difference had no statistical significance (χ2=0.215,P >0.05). Compared with water soluble eosin group similar between the two groups was statistically significant(χ2=5.221~6.923,all P <0.05)。 Microscopically, the three groups of staining sections showed clear structure and clear nucleo-cytoplasmic contrast without obvious differences. The preparation of eosin acetate solution was simpler and shorter than that of eosin hydrochloride solution.Conclusion The predyeing effect of acetic acid yihong solution on small biopsy specimens was better than that of water-soluble yihong solution, and the preparation method was better than that of hydrochloric acid yihong solution. It was easy to identify in the embedding process, and could improve the embedding speed and quality, which was worthy of promotion.
