摘要:
目的 探讨鼻咽癌患者血清miR-144-3p及miR-151-3p的表达水平及临床价值。方法 选取2016年1月~2020年7月涿州市医院收治的106例鼻咽癌患者和50例健康对照组作为研究对象,采用实时定量PCR法检测miR-144-3p及miR-151-3p表达水平。应用受试者工作特征(ROC)曲线分析miR-144-3p及miR-151-3p诊断鼻咽癌的价值。Pearson相关分析鼻咽癌患者血清miR-144-3p与miR-151-3p表达水平的相关性。结果 鼻咽癌组血清miR-144-3p(2.36±1.05 vs 0.82±0.27)及miR-151-3p(3.58±1.73 vs 0.97±0.36)表达水平均明显高于对照组,差异有统计学意义(均P <0.001)。鼻咽癌患者血清miR-144-3p及miR-151-3p表达水平升高与临床分期高、分化程度低、淋巴结转移及远处转移有关(t=5.824~19.307,均P <0.01)。ROC曲线显示, miR-144-3p为1.61及miR-151-3p为2.25是诊断鼻咽癌的最佳截断值,二者联合诊断鼻咽癌的曲线下面积(0.937,95%CI:0.875~0.99)最大,其敏感度和特异度为95.3%和87.0%。鼻咽癌患者血清miR-144-3p表达水平与miR-151-3p呈正相关(r=0.872,P<0.001)。结论 鼻咽癌患者血清miR-144-3p及miR-151-3表达水平明显升高,二者联合诊断鼻咽癌的价值较高。
Abstract:
Objective To investigate the expression level and clinical value of miR-144-3p and miR-151-3p in patients withnasopharyngeal carcinoma. Methods The 106 patients with nasopharyngeal carcinoma and 50 healthy controls who wereadmitted to Zhuozhou City Hospital from January 2016 to July 2020 were selected as the research subjects. The expressionlevels of miR-144-3p and miR-151-3p were detected by real-time quantitative PCR. The levls of miR-144-3p and miR-151-3pin the patients was analyzed by ROC curve in the diagnosis of nasopharyngeal carcinoma. Pearson correlation analysis was usedto analyze the correlation between serum miR-144-3p expression level and miR-151-3p level in patients with nasopharyngealcarcinoma. Results The expression levels of serum miR-144-3p(2.36±1.05 vs 0.82±0.27) and miR-151-3p(3.58±1.73vs 0.97±0.36) in nasopharyngeal carcinoma group were significantly higher than those in control group, the differences werestatistically significant (all P< 0.001). The increased expression levels of serum miR-144-3p and miR-151-3p were associatedwith high clinical stage, low differentiation, lymph node metastasis and distant metastasis (t=5.824~19.307, all P< 0.01). ROCcurve showed that the best cut-off values of miR-144-3p and miR-151-3p in the diagnosis of nasopharyngeal carcinoma were 1.61and 2.25, respectively. The area under the curve (0.937, 95% CI: 0.875~0.99) was the largest for the two combined diagnosis ofnasopharyngeal carcinoma, and the sensitivity and specificity were 95.3% and 87.0%, respectively. The expression levels of serummiR-144-3p was positively correlated with miR-151-3p (r=0.872, P< 0.001). Conclusion The expression levels of serum miR-144-3p and miR-151-3 in nasopharyngeal carcinoma patients were significantly increased, and the two combined diagnosis value ofnasopharyngeal carcinoma was relatively high.
