摘要:
目的 探讨支原体采用固体联合液体培养的最优组合方案。 方法 收集2016年3月~6月泌尿生殖道标本961例,采用固体(培养基 )与液体(培养液)平行培养支原体,48 h后判读结果,筛选假阳性培养液,取20 μl转种固 体培养,48 h后再次判读结果。统计分析数据,对比固体与液体不同组合培养方案的优缺点 。结果 原代培养中固体培养阳性率UU为38.7%(372/961),MH为7. 8%(75/961);液体培养阳性率UU为45.27%(435/961),MH为7.08%(68/961)。两种方法培养 UU差异率为9.47%(91/961),差异有统计学意义(χ2=43.61,P<0.05);MH差异率 为3.02%(29/961),差异无统计学意义(χ2=1.24,P>0.05)。转种固体培养与原 代固体培养差异率为53.5%(46/86)。总体(原代+转种)固体培养阳性率UU为41.9%(403/96 1),MH为8.6%(83/961);与单次(原代或转种)固体培养相比,敏感度高。总体固体培养与 单独液体培养的差异率UU为6.24%(60/961),MH为3.43%(33/961),差异有统计学意义(UU :χ2=17.067,P<0.05;MH:χ2=5.94,P<0.05)。结论 〖H T5”K〗支原体采用“两种方法平行原代培养,继而假阳性转种固体培养,以总体固体培养 为最终结果”的培养方案,兼顾固体培养特异度高的优点与液体培养敏感度高的优点,可以 提高培养准确度。
Abstract:
Objective To investigate the best experimental scheme of Mycoplasma solid culture combined with liquid culture. Methods A total of 961 samples of urogenital tract excretion were co llected from March 2016 to June 2016.Both solid culture and liquid culture were performed for detection of Mycoplasma,false positive broths were picked ou t after 48 h,20 μl of each one was transformed to solid media for subculture,final re sults were read after 48 h.The experimental data was analyzed to find an optima l combination culture scheme.Results The positive rate o f solid culture was 38.7% (372/961) for UU and 7.8% (75/961) for MH.The posit ive rate of liquid culture was 45.27% (435/961) for UU and 7.08% (68/961) for MH.The different rate between both methods was 9.47% (91/961) for UU (χ2= 43.61,P<0.005),and 3.02% (29/961) for MH (χ2=1.24,P>0.05).The different rate was 53.5% (46/86) between primary solid culture and subculture .The positive rate of total (primary solid culture and subculture) solid cultur e was 41.9% (403/961) for UU and 8.6% (83/961) for MH,which produced higher s ensitivity than single (primary solid culture or subculture) solid culture.The different rate between the total solid culture and the liquid method was 6.24% (60/961) for UU (χ2=17.067,P<0.05),and 3.43% (33/961) for MH (χ 2=5.94,P<0.05).Conclusion Perform both solid cultur e and liquid culture for Mycoplasma,pick out those false positive broths fo r subculture.Total solid culture could be more reliable as final result,for wh ich could combine specificity of solid culture with sensitivity of liquid cultur e.
