摘要:
目的 了解miR-125a/b-5p在子痫前期患者外周血血清、尿液中的表达,探讨外周循环miR-125a/b-5p在子痫前期发病中的作用。方法收集子痫前期患者(n=20)及正常妊娠组(n=20)的外周血血清及尿液,miR-125a/b-5p的表达采用基于茎环的qRT-PCR检测,同时结合临床资料进行分析。结果 血清miR-125b-5p在子痫前期组中的表达与正常妊娠组比较显著降低,差异有统计学意义(Z=-2.272,P=0.023); 血清miR-125a-5p在两组间比较,差异无统计学意义(Z=-0.622,P=0.547); 尿液miR-125a/b-5p在两组间比较,差异均无统计学意义(Z=-0.663,-0.189,P>0.05); 在子痫前期患者中,舒张压与尿miR-125b-5p存在负相关(r=-0.513,P=0.021); 正常妊娠者中,尿素氮与血清miR-125b-5p呈正相关(r=0.472,P=0.036); 收缩压与尿液miR-125b-5p,尿液miR-125a-5p呈正相关(r=0.526,0.502,P=0.017,0.024)。结论 血清miR-125b-5p的降低与子痫前期的发病相关。
Abstract:
Abstract:Objective To detect miR-125a/b-5p expressionin serum and urine of patients with preeclampsia,explore the role of miR-125a/b-5p in peripheral circulation on the pathogenesis of preeclampsia.Methods Samples of serum and urine of patients with preeclampsia(n=20),and the normal pregnancy group(n=20)were collected and the expression ofmiR-125a/b-5p was detected by stem-loop-based real-time fluorescence quantitative reverse transcriptase polymerase chain reaction.Results The expression of miR-125b-5p in the serum of patients with preeclampsia were significantly decreased compared with the normal pregnancy(Z=-2.272,P=0.023).There were no statistically differences of the expression of miR-125a-5p in serum between the two groups(Z=-0.622,P=0.547).The differences of urinary miR-125a/b-5p between patients with preeclampsia and normalpregnancy were not statistically significant(Z=-0.663,-0.189,P>0.05).Urinary miR-125b-5p was negatively correlated with diastolic blood pressurein preeclampsia group(r=-0.513,P=0.021).Serum miR-125b-5p was positively correlated with urea nitrogen in normal pregnancy group(r=0.472,P=0.036).Urinary miR-125a/b-5p were positively correlated with systolic blood pressure in normal pregnancy group(r=0.526,0.502,P=0.017,0.024).Conclusion The decrease of serum miR-125b-5p was associated with the pathogenesis of preeclampsia.
