摘要:
目的 探究CD36在隐球菌脑膜炎患者外周血单个核细胞(PBMCs)中的表达水平及其临床意义。方法 以2013年9月~2017年2月于上海长海医院和长征医院确诊的36例隐球菌脑膜炎患者外周血为实验组,以同期体检的36例健康者外周血作为对照组,采用密度梯度离心法进行PBMCs的分离,以qRT-PCR法检测PBMCs中CD36,TLR4的相对mRNA表达水平,以ELISA法检测血浆中TNF-α等细胞因子的表达水平,实验组与对照组检测值的比较采用两独立样本t检验,对实验组PBMCs中CD36的mRNA相对表达水平与TNF-α等细胞因子进行Pearson相关性分析。结果 实验组PBMCs中CD36的mRNA相对表达水平为2.01±0.63,对照组为1.49±0.47,两组比较差异具有统计学意义(t=3.969,P=0.000 2)。Pearson分析结果显示实验组CD36与血浆中IFN-γ蛋白表达水平呈正相关(r=0.384,P<0.05),与TGF-β呈负相关(r=-0.487,P<0.005)。结论 CD36可能通过影响IFN-γ,TGF-β等因子的表达参与隐球菌脑膜炎的免疫病理过程,是该病监测和治疗潜在的靶点。
Abstract:
Abstract:Objective To explore the expression level of CD36 in peripheral blood mononuclear cells(PBMCs)from patients with crytococcal meningitis and its clinical significance.Methods The experimental group was the peripheral blood samples from 36 patients diagnosed as crytococcal meningitis in Changhai Hospital and Changzheng Hospital in Shanghai from September 2013 to February 2017,while the control group was the peripheralblood samples from 36 health individuals examined at the same time.PBMCs wereseparated by density gradient centrifugation method,the relative mRNA expression of CD36,TLR4 in PBMCs was tested by qRT-PCR,the expression level of cytokines such as TNF-α in plasma was tested by ELISA,the comparison of the examinedvalues of experimental and control groups performed by two independent samples' t test,Pearson correlation analysis was performed on the relative mRNA expression of CD36 in PBMCs and cytokines such as TNF-α in experimental group.Results The relative mRNA expression of CD36 in PBMCs ofexperimental group was 2.01±0.63,the control was 1.49±0.47,and there was statistical difference(t=3.969,P=0.000 2).Pearson analysis results showed that in experimental group CD36 was positively related with theprotein expression level of IFN-γ in plasma(r=0.384,P<0.05)and negatively related with TGF-β(r=-0.487,P<0.005).Conclusion CD36 might involve in the immunopathology process of crytococcal meningitic by influencing the expression of cytokines such as IFN-γ,TGF-β,and wasthe potential target for monitoring and treating this disease.
