摘要:
目的 探讨钙调蛋白依赖性激酶Ⅱγ(CaMKⅡγ)在慢性粒细胞白血病患者从慢性期到急变期过渡的表达及其分子机制。方法 59例来自不同时期慢性粒细胞白血病患者的骨髓,通过Ficoll分离液分离单个核细胞,裂解蛋白进行Western blot检测CaMKⅡγ的表达,并初步探讨其分子机制。结果 CaMKⅡγ在慢粒加速和急变期患者中高表达,而慢性期患者表达量低或不表达,其表达与β-catenin基本一致。结论 CaMKⅡγ在慢性粒细胞白血病慢性期到加速期过渡中发挥重要作用,可通过调控β-catenin的表达影响白血病干细胞的自我更新。
Abstract:
Abstract:Objective To investigate the expression and molecular mechanism of CaMK II gamma in CML patients transitted from chronic phaseto acute phase.Methods 59 cases of bone marrow from CMLpatients in different periods were chosed.The mononuclear cells were separatedby Ficoll separation,the protein expression of CaMK II gamma was detected by Western blot,and explored its molecular mechanism.Results The expression of CaMK II gamma in patients with CML in accelerated phase andblast crisis was high.In chronic phase patients,the expression was low or no,and the expression was consistent with beta-catenin.Conclusion CaMKII gamma played an important role in the transition period from chronic phase to acute phase,and it can affect the self-renewal of leukemia stem cell through the regulation of beta-catenin.
