摘要:
目的 探讨血清miR-150-5p和miR-155-5p对老年糖尿病肾病(DN)的诊断价值。方法 采用实时定量PCR(RT-PCR)法检测126例老年DN患者(DN组),140例2型糖尿病患者(2型糖尿病组)和65例健康体检者(对照组)的血清miR-150-5p和miR-155-5p水平,分析其对老年DN患者的诊断价值。结果 DN组血清miR-150-5p(3.35±1.26 vs 1.04±0.63和0.79±0.25)和miR-155-5p(5.47±1.80 vs 1.60±0.95和1.28±0.86)表达水平均明显高于2型糖尿病组和对照组(F/χ2=8.214,10.327,均P<0.01)。2型糖尿病组与对照组血清miR-150-5p和miR-155-5p表达水平比较,差异无统计学意义(P>0.05)。ROC曲线分析显示,血清miR-150-5p和miR-155-5p诊断老年DN的最佳截值分别为2.65,4.16,二者联合诊断老年DN的AUC(95%CI)为0.892(0.834~0.953),明显优于miR-150-5p[0.805(0.749~0.865)]和miR-155-5p[0.837(0.779~0.894)],Z=7.215,4.692,P<0.05。其敏感度(90.4%)和特异度(85.7%)最好。结论 血清miR-150-5p和miR-155-5p水平在老年DN患者中明显升高,有望作为老年DN诊断的生物学标志物。
Abstract:
Objective To investigate the diagnostic value of serum miR-150-5p and miR-155-5p in elderly diabetic nephropathy(DN).Methods The serum miR-150-5p and miR-155-5p levels of 126elderly DN patients(group DN),140 patients with type 2 diabetes mellitus(type 2 diabetes group)and 65 healthy persons(control group)were detected by real-time quantitative PCR(RT-PCR),and analyzed its diagnostic value for the elderly patients with DN.Results The expression levels of serum miR-150-5p(3.35±1.26 vs 1.04±0.63and 0.79±0.25)and miR-155-5p(5.47±1.80 vs 1.60±0.95 and 1.28±0.86)in group DN were significantly higher than those in type 2 diabetes group and control group(F/χ2=8.214,10.327,all P<0.01).There was no significant difference in serum miR-150-5p and miR-155-5p levels between type 2diabetes group and control group(P>0.05).The ROC curve analysis showed that the best cut-off values of serum miR-150-5p and miR-155-5p diagnosis ofelderly DN were 2.65 and 4.16,respectively.The two combined diagnosis of elderly DN of AUC(95%CI)[0.892(0.834~0.953)] were significantly better thanmiR-150-5p[0.805(0.749~0.865)] and miR-155-5p [0.837(0.779~0.894)](Z=7.215, 4.692,P<0.05),and its sensitivity(90.4%)and specificity(85.7%)were best.Conclusion Serum miR-150-5p and miR-155-5p levels in elderly DN patients increased significantly,and it is expectedto be a biomarker for the diagnosis of DN in elderly patients.
