摘要:
目的分析骨形态发生蛋白-7 (BMP-7)在胃癌中的表达，初步探讨BMP-7对胃癌诊断的意义。方法以27例胃癌及相应的癌旁组织为研究对象，采用实时荧光定量PCR和Western blotting方法，分别检测组织中BMP-7 mRNA及其蛋白的表达情况。结果27例配对的胃癌及其癌旁组织中，63.0％(17/27)的癌组织BMP-7 mRNA的表达高于癌旁组织，差异有统计学意义(t=2.94，P<0.01，配对t检验)；Western blotting结果显示，77.8%(7/9)的癌组织BMP-7蛋白水平的表达高于癌旁组织，差异有统计学意义(t=2.57，P<0.05，配对t检验)。结论胃癌组织中BMP-7的表达高于癌旁组织，可能与胃癌的发生、发展相关，对临床胃癌的辅助诊断可能具有潜在价值。
Abstract:
ObjectiveTo preliminarily investigate the expression of BMP-7 in gastric cancer and its significance for gastric cancer diagnosis．MethodsThe expression level of BMP-7 mRNA in 27 pair-matched gastric neoplastic and adjacent non-neoplastic tissues was determined by quantitative reverse transcriptase polymerase chain reaction，and western blotting was performed to analyze BMP-7 protein level in randomly selected 9 pair-matched tissues．ResultsThe relative expression level of BMP-7 mRNA in 63.0% (17/27) gastric cancer tissues was significantly higher than that in adjacent non-neoplastic tissues (t=2.57，P<0.05，paired t-test)．The level of BMP-7 protein in 77.8% (7/9) gastric cancer tissues was higher than that in adjacent non-neoplastic tissues (t=2.57，P<0.05，paired t-test)．ConclusionCompared to the non-neoplastic tissues，expression of BMP-7 was increased in gastric cancer，indicating BMP-7 might be associated with the occurrence and development of gastric cancer，and may have potential value for the diagnosis of gastric cancer．
