摘要:
目的 研究 56℃ 30 min 热灭活后的新型冠状病毒肺炎 (COVID-19) 咽拭子及提取的核酸在 4℃保存的稳定性。方法 采用多重荧光定量 PCR 方法对一例 COVID-19 患者的咽拭子样本,56℃ 30 min 热灭活后进行 COVID-19 双靶点(ORF1ab 和 N 基因 ) 核酸检测,把原始咽拭子样本及提取的核酸保存在 4℃,分别于 24,48,72 和 96h 再次检测咽拭子样本及提取核酸中新冠病毒核酸的含量(Ct 值),分析热灭活后咽拭子样本及提取核酸在 4℃保存的稳定性。结果COVID-19 患者的咽拭子样本,56℃ 30 min 热灭活后核酸检测结果为 ORF1ab 和 N 基因双阳性,Ct 值分别为 31.27 和30.64,咽拭子样本 4℃保存 24,48,72 和 96h 后再次检测 ORF1ab 和 N 基因的结果为 24h (31.41 和 30.68),48h (34.13和 34.28),72h (34.06 和 34.11) 和 96h (36.22 和 40.02)。 提取的核酸 4℃保存 24,48,72 和 96h 后的检测结果为 24h (31.64和 30.06),48h (31.62 和 29.95),72h (31.72 和 29.75) 和 96h (31.36 和 30.09)。56℃ 30 min 热灭活的咽拭子样本在 4℃保存 24h 后,对 COVID-19 核酸的检测无明显影响 ( 偏倚 <1%),但 4℃保存 48~72h,ORF1ab 基因核酸降解约 7 倍,N 基因降解约 11 倍,4℃保存 96h, ORF1ab 和 N 基因核酸分别降解 30.91 倍和 666.29 倍。但是提取的核酸 4℃保存 24~96h 后,对核酸的检测结果无明显影响 ( 偏倚均小于 3%)。结论 研究结果表明 56℃ 30 min 热灭活后提取的核酸的稳定性比原始咽拭子样本好,热灭活后的咽拭子样本可在 4℃保存 24h。
