摘要:
目的 探讨慢性阻塞性肺疾病(COPD)患者外周血单个核细胞miR-21表达和血清IL-12水平的关系。方法 选取COPD患者共80例,对照组50例为健康人群,通过实时荧光定量PCR检测外周血单个核细胞miR-21表达,酶联免疫吸附法检测血清IL-12水平。结果 COPD稳定期组、急性期组和对照组间血糖、胆固醇和三酰甘油水平差异均无统计学意义(F=0.341~1.542,均P>0.05),而miR-21表达和IL-12水平比较差异有统计学意义(F=8.951~15.017,均P<0.05)。COPD稳定期组和急性期组miR-21表达和IL-12水平均比对照组高,差异有统计学意义(t=10.501~25.612,均P<0.05)。COPD急性期组比稳定期组miR-21表达和IL-12水平高,且差异有统计学意义(t=13.298~36.415,均P<0.05)。IL-12和miR-21存在显著的正相关(r=0.381~0.496,均P<0.05)。结论 miR-21可能参与调控IL-12的水平,miR-21表达与COPD的发病机制相关。
Abstract:
Objective To discuss the relationship of miR-21 expression and IL-12 level in patients with chronic obstructive pulmonarydisease.Methods Chosed 80 patients with suspected chronic obstructivepulmonary disease(COPD)and 50 cases as control group. Real-time RT-PCR was performed to determine the expression 1evels of miR-21 of peripheral blood mononuclear cells of three groups.To detect IL-12 level by ELISA method.Results There were no statistical difference among GLU,TC and TGlevel among stable phage,acute phage of COPD and control group(F=0.341~1.542,P>0.05).However miR-21 expression and IL-12 level had statisticaldifference(F=8.951~15.017,P<0.05).The expression 1evels of miR-21 of peripheral blood mononuclear cells of acute phage group were apparently higherthan stable phage,health group(P<0.05).The concentration of IL-12 in serum of acute COPD group was higher than other two groups(P<0.05),comparedwith stable COPD group and health group.IL-12 level was positiveiy related tomiR-21 expression(r=-0.381~0.496).Conclusion miR-2lmay be involved in the regulation of IL-12 expression.miR-21expression may be closely related to the pathogenesis of COPD.
