摘要:
目的 分析胃泌素肽前体(ProGRP)电化学发光法检测结果的可能影响因素,为检测准确性提供依据。方法 选取西安市第三医院2017年1月~7月呼吸科住院患者检测ProGRP,按0~50,51~100和>100 pg/ml三个浓度范围分别收集血浆及血清样本,每个浓度范围各筛选10例,共30例。室温放置0,2,4,8和24 h后对30例血浆样本分别进行检测,同时对0 h血清样本进行检测,分析不同放置时间、不同样本类型检测结果差异性以及不同样本类型检测结果的相关性; 收集同期健康查体人群吸烟成瘾者及不吸烟者血浆样本各30例,比较两组检测结果的差异性。结果 室温不同放置时间组(0,2,4,8,24 h)ProGRP检测结果(pg/ml)均值分别为76.29±48.06,75.55±47.19,75.59±47.17,75.99±47.07和74.64±46.57,组间比较差异无统计学意义(F=1.93,P=0.11); 血清和血浆不同样本类型组间检测结果(pg/ml)均值分别为99.30±42.40和99.10±42.60,组间差异无统计学意义(t=0.22,P=0.82),两组结果相关系数r=0.98; 吸烟组与不吸烟组间ProGRP检测结果(pg/ml)均值分别为34.91±12.51和36.80±12.49,组间差异无统计学意义(t=0.74,P=0.47)。结论 样本室温下放置24 h内化学发光法检测ProGRP结果稳定可靠且不受血浆或血清样本类型的干扰并有良好相关性; 吸烟与否对其检测结果无明显影响。
Abstract:
Objective To analyze the possible factors affecting the detection of ProGRP by electrochemiluminescence and provide basis for detecting accuracy.Methods The detected values of plasmaProGRP were divided into three successive concentration gradients.Patients fromJanuary to July 2017 in pneumology department were selected,and 10 cases of 30patients were screened for each concentration,whose plasma and serum samples were collected.Detected the plasma samples at the time point of 0,2,4,8 and 24 hours respectly at room temperature,the simultaneous detection serum samplesat 0 h,and compared the result of different laying time,different types of samples by t test and analysed the correlation of the two types specimens.Collected 30 serum samples of smoking addiction and nonsmokers respectly and compared the difference of the two groups with t test.Results The sample average of different time group placed at room temperature(0,2,4,8,24 hours)were 76.29±48.06,75.55±47.19,75.59±47.17,75.99±47.07 and 74.64±46.57 pg/ml respectively,and there was no statistically significant difference between the groups(F=1.93,P=0.11).The average of different sample types were 99.30±42.40 and 99.10±42.60 pg/ml respectively,and there was no statistically significant difference between the group(t=0.22,P=0.82).The correlation coefficient r=0.98.Test results between smoking group and non-smoking group average respectively were 34.91±12.51 and 36.80±12.49 pg/ml respectively,and there was no statistically significant difference between the group(t=0.74,P=0.47).Conclusion The ProGRP results were tested by chemiluminescence method within 24 hours atroom temperature without interference from plasma or serum sample types and hadgood correlation.Smoking or not had no significant effect on the test results.
