摘要:
目的 探究盐酸普萘洛尔对十二指肠溃疡 (duodenal ulcer, DU) 大鼠的保护作用及其机制。方法 35 只大鼠随机分为对照组、十二指肠溃疡模型组、低剂量盐酸普萘洛尔组、中剂量盐酸普萘洛尔组和高剂量盐酸普萘洛尔组,每组 7 只。采用半胱胺构建 DU 大鼠模型,采用 Western Blot 法测定蛋白浓度,采用苏木精 - 伊红染色法 (hematoxylin-eosin, HE) 评估各组大鼠的十二指肠黏膜损伤程度。分别比较各组间大鼠十二指肠中蛋白激酶 B(protein kinase B, Akt),胰岛素样生长因子 -1(insulin-like growth factors-1, IGF-1) 和 PTEN 基因 (gene of phosphate and tension homology deletedon chromsome ten, PTEN) 的表达,黏膜的损伤程度,超氧化物歧化酶 (superoxide dismutase, SOD),谷胱甘肽过氧化物酶 (glutathione peroxidase, GSH-Px) 和丙二醛 (malondialdehyde, MDA) 水平。结果 与对照组相比,模型组 Akt, PTEN和 IGF-1 的蛋白相对表达量显著降低。与模型组相比,不同剂量盐酸普萘洛尔组 Akt,PTEN 和 IGF-1 的蛋白相对表达量显著升高,其差异均具有统计学意义 (F=10.319,P < 0.01)。模型组的黏膜损伤程度显著高于对照组,低、中和高剂量盐酸普萘洛尔组大鼠的十二指肠黏膜损伤程度均显著低于模型组,其差异具有统计学意义 (F=3.703,P=0.025)。低、中和高剂量盐酸普萘洛尔组大鼠的 SOD 水平均高于模型组,高剂量组与模型组的差异具有统计学意义 (F=58.843,P=0.000)。低、中和高剂量盐酸普萘洛尔组大鼠的 GSH-Px 水平均高于模型组,高剂量组与模型组的差异具有统计学意义 (F=8.026,P=0.001)。低、中和高剂量盐酸普萘洛尔组大鼠的 MDA 水平均低于模型组,其差异无统计学意义 ( F=14.519,P=0.479)。结论 盐酸普萘洛尔通过 IGF-1/PTEN/Akt 信号通路对 DU 大鼠发挥保护作用,并且其作用可能与抗氧化有关。关键词:盐酸普萘洛尔;十二指肠溃疡;半胱胺;IGF-1/PTEN/Akt 信号通路
