摘要:
目的 探讨长链非编码RNA 肺腺癌转移相关转录因子1(long non-coding RNA metastasis associated transcriptionfactor 1 in lung adenocarcinoma,LncRNA MALAT1),微小核糖核酸(micro RNA, miR)-211-5p 表达水平变化与临床特征的相关性。方法 选取2022年1~6月河北省沧州中西医结合医院皮肤科收治的白癜风患者80 例为研究对象(白癜风组),同期于该院进行体检的健康者80 例为对照组。采用实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)法检测受试者血清中LncRNA MALAT1,miR-211-5p 表达水平;两组间比较采用独立样本t 检验;多组间(不同临床分期、临床分型、皮损面积及病程)比较进行单因素方差分析,组间多重比较采用SNK-q 检验;白癜风患者皮损面积及病程与血清中LncRNA MALAT1 和miR-211-5p 表达水平的相关性采用Pearson 法分析。结果 白癜风组患者血清LncRNA MALAT1(1.90±0.25)表达水平高于对照组(1.02±0.13),血清miR-211-5p(0.53±0.07)表达水平低于对照组(1.05±0.10),差异均有统计学意义(t=27.933,38.103,均P=0.000)。进展期及快速进展期血清LncRNA MALAT1 水平均高于稳定期(2.29±0.42, 2.47±0.31 vs 1.54±0.12),差异有统计学意义(t=15.000,16.766,均P < 0.001)。血清miR-211-5p 水平均低于稳定期(0.28±0.09, 0.22±0.06 vs 0.74±0.11),差异有统计学意义(t=24.748,25.217,均P < 0.001)。寻常型血清LncRNA MALAT1(2.06±0.26)表达水平高于节段型(1.50±0.16),寻常型血清miR-211-5p(0.39±0.11)表达水平低于节段型(0.86±0.21),差异有统计学意义(t=9.768,13.127,均P < 0.001)。泛发型血清LncRNA MALAT1 相对表达水平高于肢端型、局限型及散发型(2.59±0.38 vs 2.02±0.19,1.98±0.44,1.89±0.30),差异有统计学意义(t=5.559,6.038,7.375,均P < 0.001)。血清miR-211-5p 水平低于肢端型、局限型及散发型(0.17±0.04 vs 0.39±0.09,0.43±0.11,0.45±0.10),差异有统计学意义(t=7.651, 9.177,10.520,均P < 0.001)。皮损面积< 1%,1% ~ 50% 和> 50% 的白癜风患者血清LncRNA MALAT1 表达水平依次升高(1.69±0.19, 1.92±0.21, 2.56±0.26),差异有统计学意义(t=6.424,17.408, 12.312,均P < 0.001);皮损面积< 1%,1% ~ 50% 和> 50% 的白癜风患者血清miR-211-5p 表达水平依次升高(0.70±0.09, 0.41±0.10, 0.21±0.03),差异有统计学意义(t=18.972, 22.964, 9.012,均P < 0.001)。Pearson 相关性分析显示,白癜风患者血清LncRNA MALAT1 与皮损面积呈正相关(r=0.576,P < 0.001),血清miR-211-5p 与皮损面积呈负相关(r=-0.475,P < 0.001)。病程< 1 年、1 ~ 3 年和> 3 年的白癜风患者血清LncRNA MALAT1 表达水平依次降低(1.65±0.18, 1.88±0.22, 2.48±0.23),差异有统计学意义(t=5.984,20.581,13.291,均P < 0.001);病程< 1 年、1 ~ 3年、>3年的白癜风患者血清miR-211-5p表达水平依次降低(0.68±0.11, 0.53±0.10, 0.21±0.04),差异有统计学意义(t=8.352,24.942,15.170,均P < 0.001)。Pearson 相关性分析表明,血清LncRNA MALAT1 与病程呈正相关(r=0.344,P < 0.001),血清miR-211-5p 与病程呈负相关(r=-0.433,P < 0.001)。结论 白癜风患者血清中LncRNA MALAT1 表达上调,miR-211-5p 表达下调,且与皮损面积及病程等临床特征显著相关。
Abstract:
Objective To investigate the correlation between the expression levels of long non-coding RNA lung adenocarcinoma metastasis-associated transcription factor 1 (LncRNA MALAT1) and microRNA (miR)-211-5p and clinical characteristics. Methods From January to June 2022, 80 patients with vitiligo who were admitted to the Department of Dermatology, Cangzhou Hospital of Integrated Traditional Chinese and Western Medicine, Hebei Province were taken as the research objects (Vitiligo group). Meantime, 80 healthy people who underwent physical examination in the hospital were the control group.The expression levels of LncRNA MALAT1 and miR-211-5p in the serum of subjects were detected by real-time quantitative PCR and independent sample t-test was used for comparison between the two groups. One-way analysis of variance was used for comparison among multiple groups (different clinical stages, clinical types, skin lesion area and disease duration), and SNK-q test was used for multiple comparisons between groups. The correlation between the skin lesion area and the disease course of patients with vitiligo and the expression levels of LncRNA MALAT1 and miR-211-5p in serum was analyzed by Pearson method. Results The expression level of serum LncRNA MALAT1 (1.90±0.25) in vitiligo group was higher than that in control group (1.02±0.13), and the expression level of serum miR-211-5p (0.53±0.07) was lower than that in control group (1.05±0.10), and the differences were statistically significant (t=27.933, 38.103, all P=0.000). The serum levels of LncRNA MALAT1 in the progressive stage and rapid progressive stage were higher than those in the stable stage(2.29±0.42, 2.47±0.31 vs 1.54±0.12), and the differences were statistically significant (t=15.000, 16.766, all P < 0.001). The serum levels of miR-211-5p were lower than those in the stable stage(0.28±0.09, 0.22±0.06 vs 0.74±0.11), and the differences were statistically significant (t=24.748, 25.217, all P < 0.001). The expression level of LncRNA MALAT1 (2.06±0.26) in serum of vulgaris type was higher than that of segmental type (1.50±0.16), and the expression level of miR-211-5p (0.39±0.11) in serum of vulgaris type was lower than that of segmental type (0.86±0.21), and the differences were statistically significant (t=9.768,13.127,all P < 0.001). And the relative expression level of LncRNA MALAT1 in generalized type was higher than that in acral type, localized type and sporadic type(2.59±0.38 vs 2.02±0.19,1.98±0.44,1.89±0.30), and the differences were statistically significant(t=5.559, 6.038,7.375,all P < 0.001). The level of serum miR -211-5p was lower than that of acral type, localized type and sporadic type(0.17±0.04 vs 0.39±0.09,0.43±0.11,0.45±0.10), and the differences were statistically significant(t=7.651, 9.177, 10.520,all P < 0.001). The expression level of LncRNA MALAT1 in vitiligo patients with skin lesion area < 1%,1% ~ 50% and > 50% increased successively(1.69±0.19, 1.92±0.21, 2.56±0.26), and the differences were statistically significant (t=6.424, 17.408, 12.312,all P < 0.001). The expression level of serum miR-211-5p in vitiligo patients with skin lesions < 1%,1% ~ 50% and > 50% decreased successively (0.70±0.09,0.41±0.10,0.21±0.03), and the differences were statistically significant (t=18.972,22.964,9.012,all P < 0.001). Pearson correlation analysis showed that, in patients with vitiligo, serum LncRNA MALAT1 was positively correlated with skin lesion area (r=0.576, P < 0.001), and serum miR-211- 5p was negatively correlated with skin lesion area (r=-0.475, P < 0.001). The expression level of LncRNA MALAT1 in vitiligo patients with disease duration < 1 year, 1 ~ 3 years and > 3 years increased successively(1.65±0.18, 1.88±0.22, 2.48±0.23), and the differences were statistically significant (t=5.984, 20.581, 13.291, all P < 0.001).The expression level of serum miR-211- 5p decreased successively in patients with vitiligo with disease duration < 1 year, 1 ~ 3 years and > 3 years decreased in turn (0.68±0.11, 0.53±0.10, 0.21±0.04), and the differences were statistically significant (t=8.352, 24.942, 15.170, all P < 0.001). Pearson correlation analysis showed that serum LncRNA MALAT1 was positively correlated with the course of disease (r=0.344, P < 0.0001), serum miR -211-5p was negatively correlated with the course of disease (r=-0.433, P < 0.001). Conclusion The expression of LncRNA MALAT1 in the serum of patients with vitiligo was up-regulated, and the expression of miR-211-5p was down-regulated, and were obviously correlated with clinical characteristics such as skin lesion area and disease course.
