摘要:
目的 通过对肿瘤基因组图谱(the concet genome atlas,TCGA)的相关数据进行分析,以得到长链非编码 RNA (long non-coding RNA,lncRNA)及其下游靶分子,对60 岁以上急性髓系白血病 (acute myeloid leukemia, AML)人群预后影响的相关信息。方法 将TCGA 数据库中60 岁以上AML 患者样本,根据美国国立综合癌症网络 (NCCN)指南分为高危AML 和低中危AML 两组。通过R 软件中的“edgeR”筛选得出差异表达的lncRNA / miRNA / mRNA。再利用R 软件中的“survival”包对在高危AML 与低中危AML 中存在差异表达的lncRNA 进行生存分析,并 构建相互竞争的内源性RNA(ceRNA)网络。结果 根据TCGA 数据库中的 lncRNA / miRNA / mRNA 表达数据, 综合 分析得出10 个经典lncRNA(AC009154.1, AC011124.1, AC093627.2, AC144450.1, AL035691.1, AL355974.2, AL441943.2, LINC00703, LINC01612 和AC103702.2)与60 岁以上AML 患者预后具有显著的相关性,并得到了与这些lncRNA 相关 的ceRNA 网络。此外,还进行了两者之间存在差异表达的mRNAs 基因本体论(gene ontology,GO)和京都基因与基 因组百科全书(KEGG)通路分析。结论 可以通过干预这些lncRNA 的表达,影响高龄AML 患者预后,从而延缓或 控制高龄AML 患者病情的发展。
Abstract:
Abstract:Objective The relevant data of the cancer genome atlas(TCGA) were analyzed to obtain relevant information on the effect of long non-coding RNA (lncRNA) and its downstream target molecules on the prognosis of acute myeloid leakemia (AML) population over 60 years old. Methods AML patients over the age of 60 from the TCGA database were divided into two groups according to the NCCN guidelines: high risk AML and low and medium risk AML. Differentially expressed lncRNA/ miRNA/mRNA was screened by “edgeR” in R software. The survival package in R software was used to analyze the survival of lncrnas with differential expression in high-risk AML and low-medium-risk AML, and the ceRNA network was constructed. Results Based on lncRNA/miRNA/mRNA expression data in TCGA database, 10 classical lncRNA (AC009154.1, AC011124.1, AC093627.2, AC144450.1, AL035691.1, AL355974.2, AL441943.2, LINC00703, LINC01612 and AC103702.2) were found to have a significant correlation with the prognosis of AML patients over 60 years old after comprehensive analysis, and the ceRNA network related to these lncRNA was obtained. In addition, GO and KEGG pathway analysis of mRNAs with differentially expressed mRNAs were performed. Conclusion By intervening the expression of these lncRNA, the prognosis of elderly AML patients can be affected, thereby delaying or controlling the disease development of elderly AML patients.
