摘要:
目的 分析江苏宿迁地区汉族人群DNA甲基转移酶3B(DNMT3B)基因启动子区-149C>T和-579G>T单核苷酸多态性(SNP)与食管癌(EC)易感性的关系。方法 利用限制性片段长度多态性聚合酶链反应(PCR-RFLP)技术分析246例食管癌患者及240例健康对照者的DNMT3B启动子区-149C>T和-579G>T位点的基因型。结果 在-149C>T位点中,食管癌组和对照组的基因型TT与CT比较差异无统计学意义(χ2=0.089,P>0.05)。在-579G>T位点中,食管癌组和对照组的基因型TT与GT+GG比较差异无统计学意义(χ2=0.649,P>0.05)。在年龄和性别分组中,两位点在食管癌组和对照组间基因型频率差异均无统计学意义(χ2=0.0440.876,P值均>0.05)。结论 DNMT3B基因启动子区-149C>T和-579G>T与食管癌遗传易感性无相关性。
Abstract:
Abstract:Objective To investigate the association of DNAmethyltransferase 3B(DNMT3B)promoter region single nucleotide polymorphism(SNP)-149C>T and -579G>T with genetic susceptibility of esophageal cancer(EC)ofHan population in Jiangsu Suqian.Methods Genotypes of the -149C>T and -579G>T locus in DNMT3B promoter region were examined by polymerase chain reaction with restriction fragment length polymorphism(PCR-RFLP)in246 esophageal cancer patients and 240 healthy controls.Results In -149C>T site,the distributions of genotypes TT compared with CT inthe EC group and the controls were no significant difference(χ2=0.089,P>0.05).In -579G>T site,the distributions of genotypes TT compared with GT+GG in the EC group and the controls were no significant difference(χ2=0.649,P>0.05).When stratified by age and gender,there was no significant difference in the EC group and the controls in both two sites(χ2=0.044~0.876,all P>0.05).Conclusion The DNMT3B promoter region -149C>T and -579G>T were not associated with the genetic susceptibilityof esophageal cancer.
