摘要:
目的 探究转录因子7 类似物 2(transcription factor 7 like 2,TCF7L2)基因 rs11196205 多态性与 2 型糖尿病(type2 diabetes,T2DM)发病风险的相关性。方法 选取 2017~2018 年福建省泉州地区临床患者和健康体检人群 300 例作为研究对象,其中健康对照组 100 例, T2DM 组 200 例。其诊断标准参照中国人群糖尿病诊断标准。所有样本采用提取全血 DNA 后进行 PCR 扩增,对 TCF7L2 rs11196205 位点基因型进行测序。高效液相色谱法检测糖化血红蛋白(hemoglobinAlc, HbAlc)。全自动生化分析仪测定血清中 TC,TG,HDL-C,LDL-C,ApoA 和ApoB。采用SPSS18.0 软件进行数据整理和统计学分析。结果 T2DM 组 GC 基因型频率低于正常对照组(χ2 =5.053,P < 0.05),T2DM 组携带 C 等位基因的频率低于对照组(χ2=4.923,P < 0.05),差异有统计学意义。在不同基因型间对照组 GC 基因型的 HbAlc 水平低于 GG 基因型,差异有统计学意义(P =0.031),其余指标在对照组与 T2DM 组中 GC 与 GG 基因型的分布差异均无统计学意义(P>0.05)。二分类 Logistic 回归结果显示,BMI,HbAlc 和GLU 是T2DM 的独立危险因素,而 rs11196205基因型非 T2DM 的独立危险因素。结论 TCF7L2 rs11196205 基因多态性与T2DM 发病风险相关,C 等位基因可能降低 T2DM 的患病风险,原因可能与 C 等位基因的血糖水平较低有关。
